Tango Therapeutics Stock Jumps 21% Premarket After 82% Pancreatic Cancer Response Data — Phase 3 Is Next

2026-10-23

Tango Therapeutics (NASDAQ: TNGX) jumped about 21% in premarket trading on Friday, October 23, 2026, after the company reported an 82% objective response rate for its vopimetostat plus daraxonrasib combination in heavily pretreated pancreatic cancer — and confirmed a registrational Phase 3 trial will start in the first half of 2027. The data, presented at the ESMO 2026 Congress in Madrid, confirmed the striking signal first seen in June in a larger group of patients with longer follow-up.

The Direct Answer

What happened? Tango said its Phase 1/2 study of vopimetostat, an investigational PRMT5 inhibitor, combined with Revolution Medicines' RAS(ON) inhibitor daraxonrasib produced an 82% objective response rate (18 of 22 patients), a 100% disease control rate, and an 88% progression-free survival rate at nine months in patients with second- or third-line MTAP-deleted, RAS-mutant metastatic pancreatic ductal adenocarcinoma (PDAC). Median progression-free survival had not yet been reached at the August 20, 2026 data cutoff.

How did the stock react? Premarket movers data at 6:35 a.m. ET showed TNGX up 20.94% at $33.79, the second-biggest premarket gainer in the U.S. market, after closing Thursday at $27.93. The stock is already up more than 200% year to date.

What Happened in the Study?

The updated results cover 34 patients treated with vopimetostat plus daraxonrasib — 29 with PDAC and 5 with non-small cell lung cancer (NSCLC) — with 22 pancreatic cancer patients efficacy-evaluable after at least 14 weeks of follow-up. Most of those patients (13 of 22) were receiving the combination as third-line therapy, meaning two prior treatments had already failed. Median follow-up was 7.1 months.

Of the 18 responses, 11 were confirmed partial responses at the cutoff, and the company said all 7 initially unconfirmed responses were confirmed after the cutoff date. Chief Executive Malte Peters said the activity and tolerability first seen in the smaller June 2026 dataset had now been confirmed with longer follow-up and in a larger population, and that the response rate and durability surpass any published pancreatic cancer datasets, even in front-line settings, in the company's view.

A second combination also read out. Vopimetostat plus zoldonrasib, in patients with MTAP-deleted, KRAS G12D-mutant pancreatic cancer, produced a 54% response rate (19 of 35 patients, including one complete response), a 97% disease control rate and a 61% nine-month progression-free survival rate. In lung cancer, the daraxonrasib combination produced an 80% response rate in a small group of 5 patients.

Why Is It Blowing Up?

Pancreatic cancer is one of the hardest cancers to treat, and durable responses in patients who have already had multiple lines of therapy are rare. Principal investigator Alexander Spira of Virginia Cancer Specialists and NEXT Oncology said the large majority of patients remained progression-free at nine months despite being heavily pretreated, and called achieving that durability with an oral, chemotherapy-free regimen particularly meaningful. Anna Berkenblit, Chief Scientific and Medical Officer of the Pancreatic Cancer Action Network (PanCAN), said the depth and durability of responses warrant rapid evaluation in a larger, randomized Phase 3 trial.

Safety is the other reason investors paid attention. At dose level 1 — vopimetostat 200 mg plus daraxonrasib 100 mg, both once daily — there were no dose-limiting toxicities, no withdrawals due to treatment-related adverse events, and no Grade 4 or 5 treatment-related events among 25 safety-evaluable patients. The Grade 3 rate was 40%, and the most common side effects — mouth sores (48%), rash (44%) and diarrhea (28%) — were all Grade 1-2. A higher dose level was not well tolerated, with 3 dose-limiting toxicities in 2 patients, which is why the lower dose is the one moving forward.

The Phase 3 Plan

Following discussions with the U.S. Food and Drug Administration, Tango plans a global, randomized registrational trial in front-line patients with MTAP-deleted pancreatic cancer, starting in the first half of 2027. Patients will be randomized 1:1 to vopimetostat 200 mg plus daraxonrasib 100 mg once daily, or standard-of-care chemotherapy. The dual primary endpoints are response rate and progression-free survival, with overall survival as a key secondary endpoint. The company also intends to discuss pursuing approval in second-line pancreatic cancer with regulators.

Tango hosted a pancreatic-cancer-focused investor webcast at 7:30 a.m. ET on Friday to walk through the data, with the full results being presented at the ESMO Congress in Madrid, which runs October 23-27, with the poster session on October 25.

What's Next?

The caveats matter as much as the headline number. This is still early Phase 1/2 data in 22 efficacy-evaluable pancreatic cancer patients, not a randomized trial, and cross-trial comparisons are not conclusive. The Phase 3 does not start until the first half of 2027, and Tango remains a clinical-stage company with no product revenue. The next catalysts are the ESMO poster detail on October 25, any analyst reaction to Friday's webcast, and the company's third-quarter results, estimated for early November. FAQ

Why did Tango Therapeutics stock jump 21% premarket?
Because its vopimetostat plus daraxonrasib combination showed an 82% response rate and 88% nine-month progression-free survival in heavily pretreated pancreatic cancer, and the company confirmed an FDA-aligned Phase 3 trial starting in the first half of 2027.

What is vopimetostat?
It is Tango's investigational, oral, once-daily PRMT5 inhibitor, designed for cancers with MTAP deletion. In this study it was combined with Revolution Medicines' RAS(ON) inhibitors daraxonrasib and zoldonrasib.

How many patients were in the study?
Thirty-four patients were treated with the daraxonrasib combination, with 22 pancreatic cancer patients efficacy-evaluable at the August 20, 2026 cutoff; a separate zoldonrasib arm had 35 efficacy-evaluable patients.

When does the Phase 3 trial start?
Tango expects to initiate the global, randomized Phase 3 in front-line MTAP-deleted pancreatic cancer in the first half of 2027, comparing the combination against standard chemotherapy.
